Mast cells represent the backbone of immune system. Below we see the

The hallmark of COVID-19 is the cytokine storm, with high levels of interleukin-6

(IL-6), IL-1β, tumor necrosis factor alpha (TNF-α), i.e. proinflammatory cytokines,

released in excess by activated mast cells, which cause severe pneumonia and

neurological disorders, including stroke, disruption of blood brain barrier, and damage

to endothelial cells in the brain.


COVID-19, in its most aggressive form, is a hyperinflammatory syndrome, due to

both the excessive production of proinflammatory cytokines and the dysfunction

is due to a series of inflammatory immune cells and lung cells, like bronchial epithelial

cells, endothelial cells and fibroblasts.


Bronchial epithelial cells activated by SARS-CoV-2 cause an increase in pro-inflammatory

cytokines and induction of mast cell differentiation. But the inflammation can subside

Histamine and IL-1 are responsible for the onset of the pulmonary inflammatory reaction.

Furthermore, IL-1 together histamine can cause a sharp increase in IL-1 levels and,

Mast cells are granulocytes, which are white blood cells with granules

containing a number of powerful chemicals like histamine, serotonin, and

excessive response of mast cells activated by the virus. But the excessive

In this pathological environment, mast cell activation therefore causes an

excessive release of histamine, proteases, cytokines, chemokines and VEGF, 

which are involved in the inflammatory response. But, if the mast cells have

a normal and not unregulated metabolism, the stimulation caused by the

virus is counterbalanced by the release of anti-inflammatory cytokines,

wich are protective.


Furthermore, histamine increases the formation of IL-6, which is a proinflammatory

Then, there is also an increased production of reactive oxygen species, i.e. free radicals

and these also cause the production of pro-inflammatory cytokines.
